    秘密特工伊森（湯姆克魯斯 Tom Cruise 飾）隱藏了真實身份，準備和女友安心生活。這時，他突然接到了上級的電話。超市接頭時，直屬上司向他傳達了新的任務命令——營救負責監視歐文（菲利普帠默霍夫曼 Philip Seymour Hoffman 飾）但暴露被拘的林賽（凱麗拉塞爾 Keri Russell 飾）。任務中，他和黑人老搭檔和珍（李美琪Maggie Q 飾）並肩作戰，雖然成功救出林賽，但是林賽因腦部植入的小炸彈陣亡。伊森執著地追查著這起事故的起因，他通過線索，在梵蒂岡生擒歐文。返回途中，黑人老搭檔在林賽遺留的秘密信函中發現了一個驚天大秘密。而這時，他們中了埋伏，歐文被劫走。歐文抓了伊森的妻子要挾，逼其去上海尋找一個名叫“兔子腿”的東西。而伊森因為被控失職遭到拘捕，只能逃離總部。為了給死者報仇，讓罪魁禍首伏法，伊森踏上了去往上海尋找關鍵證據的兇險旅途。然而，真正的陰謀才剛剛開始……
